摘要: 微小RNA是一类非编码的小分子RNA, 通过参与基因的转录后水平表达调控, 调控机体的多种生理与病理进程。微小RNA(microRNA, miRNA)“封闭海绵”是一类可以封闭特定miRNA的物质。“封闭海绵”通过特定位点与mRNA的3′UTR区结合来发挥封闭微小RNA的作用。现有研究表明“封闭海绵”作为一种新型的研究miRNA的方法在生物体内、外功能丧失研究中均优于目前的反义寡核苷酸抑制剂和基因敲除方法。随着越来越多非编码RNA的发现, “封闭海绵”在研究其功能上有着不可或缺的作用。本研究对近年在“封闭海绵”的构建与应用方面的研究作一综述。

Abstract: MicroRNA(miRNA) is a kind of non-coding small RNA which participates in post-transcriptional expression modulation and involves in various kinds of physiological and pathologicalprocesses. miRNA sponges can sequester specific miRNA. miRNA sponges can bind to specific sites in the 3′untranslated region (UTR) of mRNA to sequester miRNA. Compared with antisense oligonucleotide inhibitors and genetic knockouts, miRNA sponges are valuable tools for miRNA loss-of-function studies in vitro and in vivo.With more and more non-coding RNAs were discovered, miRNA sponge has a indispensable role in the research of its function. This research will discuss the establishment and application of miRNA sponges.
