Application of proficiency testing/external quality assessment samples in trueness verification of electrochemiluminescence procedure

Abstract:

Objective To investigate the role of proficiency testing(PT)/external quality assessment(EQA) samples in trueness verification of electrochemiluminescence procedure. Methods The trueness of electrochemiluminescence procedure,such as free triiodothyronine(FT3)and estradiol(E2),was verified according to the Clinical and Laboratory Standards Institute(CLIS) EP15-A3 document 5×5 experimental design,using the samples of PT/EQA of the National Center for Clinical Laboratories. The experimental procedure,1 run per day for 5 d and 5 replicates per run,was performed. The outliers were calculated by Grubbs' test,and one way ANOVA test was used to calculate the repeatability variance(SR)of users,the within-laboratory variance (SWL),the overall standard error(SEx ),the standard error of target value(SERM)and the combined standard error(SEC). The m value was obtained from the T distribution table according to the degree of freedom(dfC). Finally,the verification interval was calculated. If the overall mean was within the verification interval,the trueness verification passed,and if out,the bias was calculated and compared with the laboratory-specified target bias. Results The overall mean values of FT3 and E2were within the correctness verification interval,and the trueness was verified. Conclusions The samples from PT/EQA can be used as a reference material for trueness verification of laboratory determination procedure.
