乙醛脱氢酶2基因多态性及生活习惯与胃癌易感性的相关性分析

Correlations of ALDH2 gene polymorphism and living habits with gastric cancer susceptibility

摘要: 目的

探讨乙醛脱氢酶2(ALDH2)基因多态性及不良生活习惯与胃癌易感性的相关性。

方法

采用基因芯片技术检测161例胃癌患者及150名体检正常者(正常对照组)ALDH2基因多态性,同时收集生活习惯资料(吸烟,饮酒,是否速饮速食、暴食,是否喜好热食、腌制食物、辛辣食物)。采用Logistic回归分析评价胃癌易感性的影响因素。

结果

胃癌组不良生活习惯比例均明显高于正常对照组(P<0.05)。ALDH2基因多态性中GG基因型频率明显高于GA和AA基因型(P<0.05),G等位基因频率明显高于A等位基因(P<0.05);但男、女性之间GG、GA、AA基因型频率及G、A等位基因频率差异均无统计学意义(P>0.05)。Logistic回归分析显示基因型[比值比(OR)=3.767,95%可信区间(CI)1.92~8.77]、等位基因(OR=3.200,95%CI 1.68~7.85)、不良生活习惯(OR=2.779,95%CI 0.85~5.82)和胃癌症状(OR=3.180,95%CI 0.99~6.72)为胃癌易感性的影响因素(P<0.05)。

结论

不良的生活习惯及ALDH2基因多态性与胃癌易感性有关。

Abstract:

Objective To investigate the correlations of aldehyde dehydrogenase 2(ALDH2) gene polymorphism and bad living habits with gastric cancer susceptibility. Methods By gene chip technology,161 patients with gastric cancer and 150 healthy subjects(healthy control group) were determined for ALDH2 gene polymorphism. The information on living habits(smoking,drinking,eating too fast,overeating and liking hot food,pickled food and spicy food) was collected. Logistic regression analysis was used for evaluating the factors of gastric cancer susceptibility. Results The proportion of bad living habits in gastric cancer group was higher than that in healthy control group(P<0.05). The frequency of GG genotype of ALDH2 gene polymorphism was higher than those of GA and AA genotypes(P<0.05),G allele frequency was higher than A allele frequency(P<0.05),but there was no statistical significance between males and females (P>0.05). Logistic regression analysis showed that genotypes [odds ratio(OR)=3.767,95% confidence interval(CI)1.92-8.77),alleles(OR=3.200,95% CI 1.68-7.85),bad living habits(OR=2.779,95% CI 0.85 - 5.82) and gastric cancer symptoms(OR= 3.180,95% CI 0.99-6.72) were risk factors for gastric cancer susceptibility(P<0.05). Conclusions Bad living habits and ALDH2 gene polymorphism are correlated with gastric cancer susceptibility.
