Expression of PBX1 in osteosarcoma tissues and its influence on prognosis

Abstract:

Objective To investigate the correlation of the expressions of pre-B-cell leukemia transcription factor 1(PBX1) gene and protein in osteosarcoma tissues with clinicopathologic parameters of patients and its influence on prognosis. Methods A total of 85 patients with osteosarcoma from the Second People's Hospital of Jiaozuo City from January 2013 to January 2016 were enrolled. The expressions of PBX1 protein and mRNA in osteosarcoma tissues and normal bone tissues(>5 cm from tumor edges) were determined,and the clinical data of patients were collected. The patients were followed up on the first day after surgery and were followed up for 5 years. Kaplan-Meier survival curve was used to analyze the survival status of patients with osteosarcoma. Cox proportional hazard regression model was used to analyze the influencing factors for prognosis in patients with osteosarcoma. Results The positive rate of PBX1 protein and the relative expression of mRNA in osteosarcoma tissues were higher than those in normal bone tissues(P<0.001). The positive rate of PBX1 protein and the mRNA relative expression in tumor tissues of patients with Enneking stage ⅡB-Ⅲ and osteosarcoma with distant metastasis were higher than those in Enneking stage Ⅰ-ⅡA and osteosarcoma without distant metastasis(P<0.05). Kaplan-Meier survival curve analysis showed that the mean survival time and 5-year survival rate of patients with PBX1 protein positive were lower than those of patients with PBX1 protein negative(P<0.001). The results of Cox proportional hazard regression model showed that Enneking stage ⅡB-Ⅲ,distant metastasis and PBX1 protein positive were risk factors affecting the survival of patients with osteosarcoma [hazard ratios(HR) were 2.861,2.244 and 2.361,95% confidence intervals(CI) were 1.204-6.799,1.047-4.814 and 1.258-5.265,respectively]. Conclusions The protein positive rate and mRNA expression of PBX1 in osteosarcoma tissues are increased,and they are related to Enneking stage and distant metastasis,which may be used as potential biomarkers for early diagnosis and prognosis evaluation of patients with osteosarcoma.
