Hook effect performance verification in laboratory-developed turbidimetric immunoassay systems

Abstract:

Objective To establish the method of hook effect performance verification in laboratory-developed turbidimetric immunoassay systems.Methods A total of 3 laboratory-developed turbidimetric immunoassay systems(Reagent A,Reagent B and Reagent C) were used for IgG with different levels. Heidel-berger curves were drawn to determine equilibrium points and safe ranges. The changes of absorbance(A),peak times and rising ranges in the rapid reaction period were observed to investigate the recognition parameters of hook effect.Results The equilibrium points of Heidel-berger curves of Reagent A,Reagent B and Reagent C were 49.0,42.0 and 105.0 g/L,respectively,and the safe ranges were 35.0-90.0,20.0-75.0 and 33.0-120.0 g/L,respectively. The hook effects of Reagent A,Reagent B and Reagent C can be identified in 49-350,28-70 and 42-350 g/L,respectively.Conclusions The recognition parameters of laboratory-developed turbidimetric immunoassay systems can be set up by performance verification of hook effect to ensure the accuracy and reliability of results.
