摘要:

环境和食品中的病原体严重威胁着人类生命健康,快速、准确的检测方法是高效防控病原的关键。目前,成簇规律间隔短回文重复序列(CRISPR)/成簇规律间隔短回文重复序列相关蛋白(Cas)13系统因其“附带剪切”的特性被广泛应用于病原体的检测,并被开发为一种灵敏度高、特异性强、成本低、快速便捷的新型核酸检测工具,开创了分子诊断的新时代。文章对CRISPR/Cas13系统的特点和分子机制进行介绍,综述CRISPR/Cas13技术在病原体检测中的研究进展,并对CRISPR/Cas13系统的未来发展和应用前景进行展望。

Abstract:

Pathogen in environment and food is a serious threat to human life and health. Rapid and accurate detection methods are the keys to the effective prevention and control of the epidemic. At present,clustered regularly interspaced short palindromic repeats(CRISPR)/clustered regularly interspaced short palindromic repeat-associated protein(Cas)13 system has been widely used in the detection of pathogens due to its "attached shear" property. It has been developed as a novel nucleic acid detection tool with high sensitivity,strong specificity,low cost,rapid and convenient,which opens a new era of molecular diagnosis. In this review,the characteristics and molecular mechanism of CRISPR/Cas13 system are introduced,the research progress of CRISPR/Cas13 technology in the detection of pathogens is comprehensively reviewed,and the future development and application prospect of CRISPR/Cas13 system are prospected.
