核酸适配体荧光探针法和液基夹层杯法检测结核分枝杆菌临床应用价值比较

Application of nucleic acid aptamer fluorescence probe method and liquid-based sandwich cup method in the determination of Mycobacterium tuberculosis

摘要:

目的 比较核酸适配体荧光探针法和液基夹层杯法检测结核分枝杆菌(MTB)的临床应用价值。方法 采集175例肺结核患者和80名健康体检者的痰液样本,分别采用液体培养法、固体培养法、抗酸染色镜检法、液基夹层杯法、核酸适配体荧光探针法检测MTB。以液体培养法结果为标准,评价核酸适配体荧光探针法、液基夹层杯法与液体培养法结果的一致性。结果 与液体培养法结果比较,液基夹层杯法的敏感性为78.08%、特异性为98.04%;核酸适配体荧光探针法的敏感性为54.79%、特异性为50.98%。结论 液基夹层杯法与液体培养法一致性较好,具有较好的临床诊断价值;核酸适配体荧光探针法性能尚需优化。

Abstract:

Objective To compare the clinical application values of nucleic acid aptamer fluorescence probe method and liquid-based sandwich cup method in the determination of Mycobacterium tuberculosis(MTB). Methods Sputum samples from 175 patients with pulmonary tuberculosis were collected and determined by liquid culture method,solid culture method,acid fast staining,liquid-based sandwich cup method and nucleic acid aptamer fluorescence probe method. Liquid culture method was used as the reference standard. The consistency of the results of nucleic acid aptamer fluorescence probe method,liquid-based sandwich cup method and liquid culture method was evaluated. Results Compared with the results of liquid culture method,the sensitivity of liquid-based sandwich cup method was 78.08%,and the specificity was 98.04%. The sensitivity of nucleic acid aptamer fluorescence probe method was 54.79%,and the specificity was 50.98%. Conclusions Liquid-based sandwich cup method is consistent with liquid culture method,and it has good clinical diagnostic value. Nucleic acid aptamer fluorescence probe method needs to be further optimized.
