哮喘患者L-Arg/ADMA比值与线粒体功能的关系

Relationship between L-Arg/ADMA and mitochondrial function in patients with asthma

摘要:

目的 探讨哮喘患者左旋精氨酸(L-Arg)、不对称二甲基精氨酸(ADMA)和L-Arg/ADMA比值与线粒体功能指标[血小板线粒体膜电位(MMP)、三磷酸腺苷(ATP)、细胞色素C氧化酶(COX)]的关系。方法 选取2016年11月—2017年5月新疆医科大学附属中医医院支气管哮喘急性发作期患者122例(哮喘组)和健康体检者44名(正常对照组)。检测所有研究对象MMP、ATP、COX活性和血浆L-Arg、ADMA水平,并计算L-Arg/ADMA比值。采用Pearson相关分析评估各项指标的相关性。结果 与正常对照组比较,哮喘组MMP、COX和血浆L-Arg、L-Arg/ADMA比值降低(P<0.05),ATP升高(P<0.05)。2个组之间血浆ADMA水平差异无统计学意义(P>0.05)。线粒体ATP与血浆ADMA呈负相关(r=-0.404,P<0.001),与L-Arg/ADMA比值呈正相关(r=0.238,P=0.008);线粒体COX与血浆ADMA呈负相关(r=-0.269,P=0.003)。其他指标无相关性(P>0.05)。结论 哮喘患者L-Arg水平异常,存在精氨酸代谢紊乱,且与线粒体功能异常有关。

Abstract:

Objective To investigate the relationship between the levels of L-arginine(L-Arg) and asymmetric dimethylarginine(ADMA) in asthma patients and L-Arg/ADMA ratio,as well as the mitochondrial function indicators [platelet mitochondrial membrane potential(MMP),adenosine triphosphate(ATP) and cytochrome C oxidase(COX)]. Methods Totally,122 patients with acute exacerbation of bronchial asthma(asthma group) and 44 healthy subjects(healthy control group) were enrolled from the Affiliated Hospital of Traditional Chinese Medicine of Xinjiang Medical University from November 2016 to May 2017. The activities of MMP,ATP and COX,as well as the levels of plasma L-Arg and ADMA were determined,and the L-Arg/ADMA ratio was calculated. Pearson correlation analysis was used to evaluate the correlations among the various indicators. Results Compared with healthy control group,the levels of MMP,COX and plasma L-Arg,as well as the L-Arg/ADMA ratio in asthma group were decreased(P<0.05),while the ATP level was increased(P<0.05). There was no statistical significance in plasma ADMA levels between the 2 groups(P>0.05). Mitochondrial ATP was negatively correlated with plasma ADMA(r=-0.404,P<0.001),and it was positively correlated with the L-Arg/ADMA ratio(r=0.238,P=0.008). Mitochondrial COX was negatively correlated with plasma ADMA(r=-0.269,P=0.003). There was no correlation among the other indicators(P>0.05). Conclusions The L-Arg level in asthma patients is abnormal,indicating a disorder in arginine metabolism,which is related to mitochondrial dysfunction.
