PFTK1表达与非小细胞肺癌微血管密度和患者预后的关系

Relationship between PFTK1 expression level and microvessel density and postoperative prognosis in non-small cell lung cancer

摘要:

目的 探讨PFTAIRE蛋白激酶1(PFTK1)表达与非小细胞肺癌(NSCLC)患者癌组织微血管密度(MVD)和术后预后的关系。方法 选取2013年1月—2015年1月南阳市中心医院行手术治疗的NSCLC患者88例,收集其癌组织和癌旁组织(距离肿瘤边缘>2 cm),检测PFTK1表达,并进行MVD计数。采用Spearman相关分析评估PFTK1表达与MVD计数的相关性。采用Kaplan-Meier生存曲线和Log-rank χ2检验分析NSCLC患者的生存情况。采用Cox回归分析评估NSCLC患者预后(死亡)的危险因素。结果 88例NSCLC患者中,PFTK1总阳性率为70.45%(62/88)。不同TNM分期、肿瘤直径和有无淋巴转移的NSCLC患者之间PFTK1阳性率和MVD计数差异均有统计学意义(P<0.001、P<0.05),不同性别、年龄、病理分型和组织学分级的NSCLC 患者之间PFTK1阳性率和MVD计数差异均无统计学意义(P>0.05)。PFTK1阳性组MVD计数显著高于PFTK1阴性组(P<0.05)。Spearman相关分析结果显示,PFTK1表达与MVD计数呈正相关(r=0.952,P<0.001)。多因素Cox回归分析结果显示,TNM分期Ⅲ期、有淋巴转移、肿瘤直径>3 cm、PFTK1阳性是NSCLC患者预后(死亡)的独立危险因素(P<0.05)。结论 NSCLC患者癌组织中PFTK1呈高表达,且与MVD计数和患者预后密切相关。

Abstract:

Objective To investigate the relationship between the expression of PFTAIRE protein kinase 1(PFTK1) and microvessel density(MVD) and postoperative prognosis in non-small cell lung cancer(NSCLC) patients. Methods A total of 88 NSCLC patients who underwent surgical treatment at Henan Nanyang Central Hospital from January 2013 to January 2015 were enrolled. Cancer tissues and adjacent tissues(>2 cm from tumor edges) were collected,and PFTK1 expression and MVD count were determined. Spearman correlation analysis was used to evaluate the correlation between PFTK1 expression and MVD count. Kaplan-Meier survival curve and Log rank χ2 test were used to analyze the survival status of NSCLC patients. Multivariate Cox regression analysis was used to evaluate the risk factors for prognosis(death) in NSCLC patients. Results Among the 88 NSCLC patients,the total positive rate of PFTK1 was 70.45%(62/88). There was statistical significance in PFTK1 positive rate and MVD count among NSCLC patients with different TNM stages,tumor diameters and lymph node metastasis(P<0.001,P<0.05),while there was no statistical significance in PFTK1 positive rate and MVD count among NSCLC patients with different sex,ages,pathological classification and histological grading(P>0.05). MVD count in PFTK1 positive group was higher than that in PFTK1 negative group(P<0.05). Spearman correlation analysis showed a positive correlation between PFTK1 expression and MVD count(r=0.952,P<0.001). Multivariate Cox regression analysis showed that TNM stage Ⅲ,lymph node metastasis,tumor diameter >3 cm and PFTK1 positivity were independent risk factors for prognosis(death) in NSCLC patients(P<0.05). Conclusions PFTK1 is highly expressed in cancer tissues of NSCLC patients,and it is closely related to MVD count and patient prognosis.
