骨保护素在糖尿病大鼠主动脉粥样斑块和血管钙化中的表达

Osteoprotegerin expression on aortic atherosclerotic plaques and vessel calcification of diabetes mellitus rats

摘要:

目的 探讨骨保护素(OPG)在糖尿病大鼠主动脉粥样斑块和血管钙化中的表达水平以及吡格列酮对其的干预作用。方法 选取健康雄性SD大鼠42只,随机分为6组[对照组(A组)、糖尿病组(B组)、主动脉粥样斑块和血管钙化组(C组)、糖尿病+主动脉粥样斑块和血管钙化组(D组)、主动脉粥样斑块和血管钙化+吡格列酮组(E组)、糖尿病+主动脉粥样斑块和血管钙化+吡格列酮组(F组)],每组7只。各组大鼠于实验第30天取血并分离血清,检测高敏C反应蛋白(hs-CRP)、空腹血糖(FPG)、HbA1c和OPG;同时进行胸主动脉钙沉积含量和碱性磷酸酶(ALP)活性测定。结果 糖尿病大鼠造模成功率为100%。与A组比较,C组、D组和E组胸主动脉钙沉积含量、ALP活性及血清OPG、hs-CRP水平明显升高(P<0.05);F组胸主动脉钙沉积含量及ALP活性明显升高(P<0.05),而血清hs-CRP、OPG水平差异无统计学意义(P>0.05)。C组胸主动脉钙沉积含量、ALP活性及血清OPG水平与E组比较,差异均有统计学意义(P<0.05)。D组胸主动脉钙沉积含量、ALP活性及血清hs-CRP、FPG、HbA1c、OPG水平与E组和F组比较,差异均有统计学意义(P<0.05)。F组胸主动脉钙沉积含量和血清OPG水平明显低于E组(P<0.05)。OPG与血管钙含量、ALP、hs-CRP、FPG呈正相关(r值分别为0.654、0.218、0.539、0.711,P<0.05或P<0.01)。结论 吡格列酮可降低大鼠主动脉粥样斑块和血管钙化的形成,其机制可能与调控OPG的表达,抑制炎症反应,降低钙盐沉积有关。

Abstract:

Objective To investigate the expression of osteoprotegerin(OPG) on aortic atherosclerotic plaques and vascular calcification of diabetes mellitus rats and the interference of pioglitazone. Methods A total of healthy male SD rats were enrolled and classified into 6 groups randomly,and there were 7 rats in each group,including control group (group A),diabetes mellitus group(group B),aortic atherosclerotic plaque and vessel calcification group(group C),diabetes mellitus+aortic atherosclerotic plaque and vessel calcification group(group D),aortic atherosclerotic plaque and vessel calcification+pioglitazone group(group E) ,diabetes mellitus+aortic atherosclerotic plaque and vessel calcification+pioglitazone group(group F). Blood was extracted from rats on the 30th day for the determinations of high-sensitivity C-reactive protein(hs-CRP),fasting plasma glucose(FPG),glycated hemoglobin A 1c(HbA1c)and OPG. Calcium deposits in thoracic aorta and alkaline phosphatase(ALP) activity were determined. Results The success rate of diabetes mellitus rat modeling was 100%. Compared with group A,the levels of calcium deposits in thoracic aorta,ALP activities,OPG and hs-CRP in group C,D and E increased(P<0.05). The levels of calcium deposits in thoracic aorta and ALP activities increased(P<0.05),and there was no statistical significance for hs-CRP and OPG in group F(P>0.05). The levels of calcium deposits in thoracic aorta,ALP activities and OPG had statistical significance between group C and E(P<0.05). The levels of ALP activities,hs-CRP,FPG,HbA1c and OPG between group D and F were statistically significant(P<0.05). The levels of calcium deposits in thoracic aorta and OPG in group F were lower than those in group E(P<0.05). OPG was positively related with the levels of calcium deposits in thoracic aorta,ALP activities and hs-CRP and FPG(r=0.654,0.218,0.539 and 0.711,P<0.05 or P<0.01). Conclusions Pioglitazone can reduce the formation of aortic atherosclerotic plaques and vessel calcification,and the mechanism might be related to the regulation of OPG,the inhibition of inflammatory reaction and the decreasing of calcium salt deposition.
