降低血清中脂多糖测定干扰因素的分析

摘要:

目的 优化鲎试剂动态显色法检测血清样本中脂多糖的处理工艺,降低脂多糖检测中的干扰因素。方法 在单因素试验基础上,采用三因素三水平的响应面分析法研究表面活性剂用量、碱用量、处理时间及其交互作用对样本中脂多糖检测结果的影响。结果 最佳样本处理工艺为:0.6% Triton X-100与0.020 mol/L KOH等体积混匀作为样本处理液,处理时间10 min。在该工艺下,干扰因素最少,临床符合率可达83.25%。结论 建立的最佳样本处理工艺对临床特殊样本(黄疸、溶血、乳糜)的抗干扰能力强,优化后的脂多糖测定方法可显著提高临床样本检测的准确率。

Abstract:

Objective To optimize treatment process of chromogenic kinetic limulus amebocyte lysate assay for lipopolysaccharide in serum and reduce interference factors.Methods On the basis of single factor experiments,response surface methodology with 3 factors and 3 levels was used to study the effects and interactions of surfactant,alkali dosage and treatment time.Results The treatment process was optimized as follows:0.6% Triton X-100 mixed with equal volume of 0.020 mol/L KOH as treatment reagent,treatment time 10 min. Under these conditions,the clinical coincidence rate was 83.25%. Conclusions This treatment process can be used to detect special samples,including jaundice,hemolysis and chyle,and can provide a reference for clinical diagnosis.
