口腔白念珠菌耐药性及毒力因子表达水平相关性研究

Correlations of virulence factor expression and drug resistance for Candida albicans isolated from oral cavity

摘要:

目的 了解口腔白念珠菌分泌型水解酶、生物膜和菌丝相等毒力因子的体外表达水平及其耐药性,并分析两者的相关性。方法 分别采用牛血清白蛋白平板、卵黄琼脂平板和三丁酸甘油脂平板检测口腔白念珠菌分泌型天冬氨酸蛋白酶(SAP)、磷脂酶(PL)和脂肪酶(Lip)的活性;采用结晶紫法检测其生物膜的形成,采用实时荧光聚合酶链反应(PCR)检测菌丝壁蛋白1(HWP1)基因的表达;检测白念珠菌对常用抗真菌药物的耐药性。结果 80%的菌株高产分泌型SAP,98%的菌株高产PL,78%的菌株Lip产量中等,无菌株高产Lip;50株菌株均能形成生物膜,其中30株(60%)生物膜形成能力强;SAP和PL的体外表达水平呈正相关(P<0.05),其他毒力因子之间均无相关性;耐药株与敏感株的毒力因子体外表达及HWP1表达之间没有显著性差异。结论 大部分口腔白念珠菌高表达SAP和PL,中等表达Lip,生物膜形成能力强,SAP和PL的体外表达水平呈正相关,口腔白念珠菌的毒力与耐药性之间没有显著相关性。

Abstract:

Objective To study the in vitro expression of virulence factors and the drug resistance of oral Candida albicans,including the secretion of hydrolytic enzymes,biofilm formation and mycelium phase production,and to investigate the correlations. Methods The activities of secreted aspartyl proteinase(SAP),phospholipase(PL) and lipase(Lip) of Candida albicans were determined by bovine serum albumin agar,egg yolk agar and tributyrate agar,respectively. The formation of biofilm was determined by crystal violet staining assay,and the expression of hyphal wall protein 1(HWP1) gene was determined by real-time fluorescence polymerase chain reaction(PCR). Antibiotic susceptibility test was performed. Results The 80% isolates produced a high level of SAP,98% isolates displayed a high level of PL,78% isolates produced a medium level of Lip,and none displayed a high level of Lip. All isolates were able to form biofilm,and the ability of biofilm formation of 30 isolates(60%) was strong. There was a positive correlation between the in vitro expressions of SAP and PL(P<0.05). No relationship was found in the other virulence factors,while there was no statistical significance in the level of virulence factors in vitro and HWP1 expression between drug-resistant and drug-sensitive isolates. Conclusions Most of isolates display high levels of SAP and PL and medium levels of Lip,which produce biofilm strongly. There is a positive correlation between the in vitro expressions of SAP and PL. The virulence of oral Candida albicans has no correlation with drug resistance.
