Abstract:

Objective To establish a new type of plastic microfluidic chip which can detect Escherichia coli O157:H7 rapidly for the early diagnosis and differentiation. Methods The device contained microfluidic channels fabricated from cycloolefin polymer(COP) and optically clear adhesive(OCA). The microfluidic channels on OCA film were simply fabricated using a laser prototyping technique. Using immunofluorescence assay,Escherichia coli O157:H7 was detected. The consistency between microfluidic chip and culturing was evaluated. Results A new and simple microfluidic chip system had been established successfully. This system had a low detection limit of 103 CFU/mL,the repeatability and specificity were good, and the overall consistency with culturing was 95%. Conclusions This new type of COP plastic microfluidic chip system could detect Escherichia coli O157:H7 rapidly and inexpensively.
