不同分期宫颈癌患者外周血Treg和相关细胞因子差异分析

Regulator T cells and cytokines in peripheral blood of cervical cancer patients with different stages

摘要:

目的探讨不同分期宫颈癌患者外周血调节性T细胞(Treg)比例和相关细胞因子的变化规律。方法 将80例不同分期宫颈癌患者和24名健康女性纳入研究,使用酶联免疫吸附试验(ELISA)检测血清白细胞介素(IL)2、IL-10、转化生长因子β(TGF-β)、细胞毒T淋巴细胞相关抗原4(CTLA-4)和淋巴细胞活化基因3(LAG-3)水平,采用流式细胞术检测外周血Treg比例。结果 宫颈癌患者血清IL-2水平显著低于对照组(P<0.01),且随分期升高而降低;IL-10和TGF-β水平高于对照组(P<0.05),且随着分期升高而升高;CTLA-4水平高于对照组(P<0.05),但不同分期间差异无统计学意义(P>0.05)。宫颈癌Ⅲ期患者LAG-3水平高于对照组(P<0.05)。宫颈癌患者外周血Treg比例显著高于对照组(P<0.05),患者Treg比例随分期升高而增加。结论 外周血IL-2、IL-10和TGF-β水平及Treg比例与宫颈癌的发展存在密切联系,对这类指标的监测和分析有利于揭示宫颈癌发展的免疫机制。

Abstract:

Objective To investigate regulator T cell(Treg) ratio and cytokines in peripheral blood of cervical cancer patients with different stages. Methods A total of 80 cervical cancer patients with different stages and 24 healthy females were enrolled. The levels of interleukin(IL)-2,IL-10,transforming growth factor-beta(TGF-β),cytotoxic T-lymphocyte antigen 4(CTLA-4) and lymphocyte activation gene 3(LAG-3) in serum were determined by enzyme-linked immunosorbent assay(ELISA),and flow cytometry was used to determine the ratio of Treg in peripheral blood. Results The level of IL-2 in patients with cervical cancer was lower than that in healthy control group(P<0.01),and decreased with stage increasing. The levels of IL-10 and TGF-β were higher than those in healthy control group(P<0.05),and increased with stage increasing. The level of CTLA-4 was higher than that in healthy control group(P<0.05),but there was no statistical significance among different stages(P>0.05). The level of LAG-3 at stage Ⅲ was higher than that in healthy control group(P<0.05). The ratio of Treg in cervical cancer patients was higher than that in healthy control group(P<0.05), and increased with stage increasing. Conclusions The levels of IL-2,IL-10 and TGF-β and Treg ratio in peripheral blood are closely related to the development of cervical cancer. The monitoring and analysis of these indices will help to reveal the immune mechanism of cervical cancer.
